Hi Chipie, and welcome to the forum. I am assuming that you are in the US as bio-spira is not available in the UK.
When you do water changes, you always have to treat the water with a water conditioner - as this takes the chlorine out of the water. Chlorine will kill the useful bacteria which has built up in the filter.
You should check with your local water authority to see if chloramines are added to the water, and if so, you will require a conditioner which removes both chlorine and chloramines. I think Tetra Aquasafe in the US does this.
If you are using the bio-spira to cycle the tank and have no fish yet, then I don't think you should be doing water changes, but I would check that if I were you as I've never used it before.
